A histological evaluation of cellular response on bone regeneration scaffold

Biocompatibility testing for a new implant material is necessary before clinical use. The materials bone scaffolding must also go through biocompatibility tests both in vitro and vivo. purpose of this study was to compare different staining methods histological preparations assess the tissues. Twenty Wistar rats (n= 20), aged 4 months, males, weighing about 250-350 g were divided into 2 research groups, namely sub-cutaneous implantation 3, 14, 30 days. sample biodegradable with gypsum calcium carbonate as basic ingredients. Histological stains prepared several ways using Hematoxylin Eosin (HE), Mallory, Toluidine Blue, Tartrate-resistant acid phosphatase polyclonal antibody (TRAP), Chromogen 3,3 diaminobenzidine (DAB) after pre-performing decalcification process ethylenedinitril-o-tetra-acetic (EDTA). observation done magnification ranging from 10X10 100X10 light microscope (Nikon eclipse E600) OptiLab viewer. results displayed form descriptions images generated methods. Staining by HE method most often chosen because it cheap easy but other such TRAP show better contrast. Tissue techniques immunohistochemistry (DAB, VEGF) are used evaluate cell tissue responses identifying specific proteins present.
